After working in theatre, she made her feature film acting debut in the two-part crime drama *Gangs of Wasseypur* (2012), earning nominations for the Filmfare Award for Best Female Debut and Best Supporting Actress. Her subsequent releases included *Ek Thi Daayan* (2013), *D-Day* (2013), *Dedh Ishqiya* (2014), *Badlapur* (2015), and the comedy *Jolly LLB 2* (2017). During this period, she also made her Marathi debut in *Highway* (2015), her Malayalam debut in *White* (2016), and appeared in *Viceroy's House* (2017). In Tamil cinema, she appeared in *Kaala* (2018) and *Valimai* (2022). In streaming, Qureshi starred in the series *Leila* (2019), *Maharani* (2021–present), and *Mithya* (2022–2024). She made her international film debut in *Army of the Dead* (2021), followed by *Monica, O My Darling* (2022), and portrayed the titular chef in the biographical drama *Tarla* (2023). Her later film credits include *Bayaan* (2025), *Maalik* (2025), *Single Salma* (2025), *Jolly LLB 3* (2025), and *Gulabi* (2026).
